Start -> Control Panel -> System -> Hardware Tab -> Device Manager

Check to see if any devices have a yellow exclamtion mark next to them.

Click the + next to the Sound, video and game controllers.

Check that your sound card is listed here, or if you not sure post to tell us what is in the list. You double click your sound card and it will show the properties and have a Device Status, which says if it is working. You can reinstall the device drivers from here if necessary.

Just in case:
Have you restarted your PC and checked the speakers are working and plugged in.
