Ok so say hypothetically this friend of mine met this nice laydee online and they chatted and stuff, swapped pics and even the odd phone call. And suppose this friend of mine started having minor niggling doubts about, say, whether the other person was entirely naturally born with the gender with which they were now presenting themselves, (you know like when you have a queen in your hand and you're convinced you're holding the nuts), how would one politely go about ascertaining for sure the gender identity of one's laydee without causing gross offence?

If you were just chatting etc would it even be an issue at that point?  I shouldn't worry about anything like that until after you've met up and decided if you want to take things further.  If you were just going to be friends, is her birth gender of any relevance?  I'm sure if things did start getting more serious and this woman had had gender reassignment at some point she would raise the issue with you (I mean your friend ) then.

For more super sleuth tactics, perhaps your friend could send a picture of when he was a little boy, and see if the woman responds by sending her own picture?  Friends reunited might be somewhere to look too.

I think "issues being raised" is chief amongst my worries. It's a difficult subject 2 tackle. I know some people like to wait for a relationship to mature but I think there are some things to get 'out in the open' right away.
From the profile "I have never married or had kids". I think this is maybe due to lack of ovaries, normally a prerequisite.
